چکیده

Lapachol whose isolation and study at the end of the century by Parternó, is one of the most brilliant chapters of Phytochemistry, especially in conjunction with the work of Max Siewert a communication on the wood of the Argentina Lapachol (Bignoniaceae) tree from Max Siewert, submitted along with Ricardo Napp in Exp. NAC. Philadelphia in 1876. The Lapachol has its origin in the Tabebuia avellanedae Lorentz ex. Griseb (Bignoniaceae) which in Brazil is known as Pau D Arc, Ipê-purple, Ipê-Black, and Yellow-Ipê. It is a tree of huge size used in traditional medicine for cases of stomatitis, inflammatory processes in general and against cancer. -Lapachone is an o-naphthoquinone from Lapachol used against, Trypanosome cruzi, cancer, viruses, bacteria, and leishmania is currently evaluated. This plant can also be found in other plant families such as, Verbenaceae Leguminosae, Sapotaceae, Scrophulariaceae, Malvaceae and Proteaceae. This paper aims at demonstrating the pharmacology and clinical activity of Lapachol and -Lapachone, through their mechanism of action, and other actions which are not related to their anti neoplastic activity.

Chemical reactivity studies with naphthoquinones from Tabebuia with anti-trypanosomal efficacy.

The biological activities of the naphthoquinones lapachol and its cyclization product beta-lapachone, extracted from trees of the genus Tabebuia, have been intensively studied. Given continuity to the studies about heterocyclic derivatives obtained from the reaction of these naphtoquinones with amino-containing reagents, 22 derivatives of beta-lapachone, nor-beta-lapachone and lapachol were syn...

Microbial transformations of natural antitumor agents: conversion of lapachol to dehydro-alpha-lapachone by Curvularia lunata.

Microbial transformation of lapachol, a naturally occurring naphthoquinone, was carried out by Curvularia lunata (NRRL 2178). The fungus brings about oxidative cyclization of the substrate to dehydro-alpha-lapachone, which was isolated and characterized by nuclear magnetic resonance and mass spectral analyses; its structure was verified by chemical synthesis. The metabolite is a naturally occur...

The trypanocidal activity of naphthoquinones: a review.

Naphthoquinones are compounds present in several families of higher plants. Their molecular structures confer redox properties, and they are involved in multiple biological oxidative processes. In folk medicine, especially among Indian populations, plants containing naphthoquinones have been employed for the treatment of various diseases. The biological redox cycle of quinones can be initiated ...

Studies on novel polycyclic heterocycles: synthesis of new naphthaquinoxaline and naphthazaquinoxaline derivatives from naturally occurring quinones

A facile synthesis of novel polycyclic heterocycles namely naphthaquinoxaline and naphthazaquinoxaline derivatives is carried out by the reaction of lapachol and β-lapachone, naturally occurring naphthoquinones with o-phenylene diamine and 2,3-diaminopyridine. Regioselectivity in the reaction of β-lapachone with 2,3-diaminopyridine is confirmed by single crystal X-ray diffraction of a represent...

ANTHELMINTIC ACTIVITY OF LAPACHOL, β-LAPACHONE AND ITS DERIVATIVES AGAINST Toxocara canis LARVAE

Anthelmintics used for intestinal helminthiasis treatment are generally effective; however, their effectiveness in tissue parasitosis (i.e. visceral toxocariasis) is moderate. The aim of this study was to evaluate the in vitro activity of lapachol, β-lapachone and phenazines in relation to the viability of Toxocara canis larvae. A concentration of 2 mg/mL (in duplicate) of the compounds was tes...
